I think Fluttershy is closest to the "camera" (or POV, if you will), that is why she appears bigger. She does have the biggest boobs though, so Xane's theory holds up! XD

I've written a little bit of dialogue to go with the pic.

Rarity: Twilight darling, please tell me you know how to fix this?
Twilight: Well, I DO know the counter spell. But... I don't have a horn anymore, casting it wil be... tricky.
Rainbow Dash: Nice job, egghead.
Pinkie Pie: Wow, Twilight! Did you know humans were so big?
Fluttershy: I - I don't think she's big, Pinkie Pie... I think... we're small... *blush*

this is suprisingly a lot like what I imagined the ponies would look like is girls

and FS is the biggest because of the shyness curse. Usually, it's those that don't want to stand out that always get the most attention grabbing features.

Saber (Genderswapped King Arthur) and Irisviel from Fate/Zero. The characters are pretty vague like this, but the city beneath them is definitely Fuyuki city (judging from Waver's maps in the anime series).
